The company has four branches but the new 50,000sq ft store will be the largest when it opens later this month.
It aims to offer everything under one roof, selling items from screwdrivers to sewing kits and all at bargain prices.
What Stores will employ between 40 and 50 staff, full and part-time.
General manager Barry Hughes said: "We sell a wide variety of choice, from needle and cotton to tools and DIY. We sell a lot of 'as seen on TV' but for much cheaper prices. It's a great opportunity in a successful retail park. We have been looking for a larger premises. In the credit crunch people are looking for value for money, and that's what we are."
The company began 15 years ago in Bristol and had been based at the Longwell Green retail park until it was redeveloped. Mr Hughes said the firm then hoped to relocate to similar premises in Bristol but couldn't find anything large enough so moved to Cardiff.
The new store will be the third of the former Woolies in Bristol to be reopened since the iconic chain fell victim to the credit crunch. A branch in Kingswood is set to reopen as a furniture and electrical store for the charity British Heart Foundation in September after a major refit.
And another, in Yate, was occupied by a budget fashion retailer and Bonmarche womenswear in June.
DIY store Proper Job is planning to occupy the former Woolies in Clevedon and discount chain Factory Outlet Stores took over the Nailsea shop in May.
